The Ampli-sRANKL ELISA is an enzyme immunoassay intended for the quantitative determination of human free soluble RANK-Ligand in Plasma, Serum and Cell Culture supernatants. sRANKL (soluble receptor activator of nuclear factor NF-kB ligand, also known as Osteoprotegerin ligand, OPGL) is a member of the tumor necrosis factor (TNF) family, and is the main stimulatory factor for the formation of mature osteoclasts. sRANKL is produced by osteoblastic lineage cells and activated T lymphocytes, and it activates its specific receptor RANK that located on osteoclasts and dendritic cells. The effects of sRANKL are counteracted by osteoprotegerin (OPG), which is secreted by various tissues and acts as an endogeneous soluble receptor antagonist. Some recent emphasis has also been placed on the potential role of OPG and sRANKL in vascular calcification. As the serum concentration of sRANKL is usually quite low in normal samples an additional enhancement system is used in this ELISA.
